Product Introduction
This kit employs a proprietary bifunctional fusion protein that specifically and covalently binds a designated single-stranded DNA (ssDNA) sequence through one functional domain, while the other domain selectively and stably binds the Fc region of a broad range of IgG antibodies. This unique design enables highly efficient antibody–ssDNA conjugation with a defined 1:1 antibody-to-ssDNA stoichiometric ratio, ensuring excellent conjugate uniformity, reproducibility, and performance in downstream applications.


Protocol
1. Preparation of the ssDNA–AND Linker Intermediate
In a 200 μL PCR tube, add the following components in the order listed:
- 10 μL Buffer (Yellow Cap)
- 3.5 μL Ultrapure Water (Clear Cap)
- 2 μL ssDNA
- 3.5 μL AND Linker (Pink Cap)
Mix thoroughly by gentle pipetting, then incubate at 37°C in a water bath or dry heating block (heat block) for 60 minutes.
2. Antibody Conjugation
Add 12 μL of the IgG antibody to be conjugated to the reaction mixture prepared in Step 1.
Mix thoroughly and incubate at 4°C for 60 minutes.
3. Purification of the IgG–ssDNA Conjugate
To remove unreacted ssDNA and excess AND Linker, purify the reaction mixture from Step 2 using either a Sephadex G-50 desalting column or an ultrafiltration centrifugal filter unit.
