In vivo fluorescence imaging places extremely high demands on deep-tissue penetration and low background signal. NIR dyes meet these demands head-on: their longer wavelengths substantially reduce light scattering and autofluorescence from biological tissue, making them the preferred tool for small-animal in vivo imaging in North American preclinical and translational research settings.
ICG (Indocyanine Green) — the only FDA-approved tricarbocyanine NIR dye — is the clinical benchmark. It peaks at ~790 nm absorption, binds rapidly to plasma proteins after intravenous injection, and is selectively taken up by the liver and excreted into bile, with a half-life of roughly 150–180 seconds. These properties underpin its widespread use in fluorescence-guided surgery, liver function assessment, and ophthalmic angiography. Its dicarboxylic acid derivative carries terminal carboxyl groups for bioconjugation and functionalized probe development.
IR783 has been reported to inhibit tumor growth and induce apoptosis, making it well suited to tumor NIR imaging and anticancer activity studies.
IR26 emits at ~1190 nm, placing it in the NIR-II window, with photostability superior to many NIR dyes — an advantage for demanding deep-tissue and long-acquisition protocols.
Cy5, a far-red cyanine dye, is widely used for antibody labeling, flow cytometry, and fluorescence-guided surgery.
For research use only unless otherwise noted. ICG is FDA-approved for specific clinical indications; other dyes listed are intended for laboratory and preclinical research.
