Innovative Deuterated Methylated Aromatics by Professor Zhou Qianghui: Transforming Medicinal Chemistry
Methyl groups are commonly found in small-molecule drugs and play a vital role in enhancing drug properties. Numerous studies demonstrate that introducing methyl groups at specific sites in candidate drugs can:
- Improve metabolic stability,
- Shorten half-life,
- Enhance solubility and target selectivity, and
- Alter drug effects, such as converting an agonist into an antagonist.
When the hydrogen atom in the methyl group is replaced with a deuterium atom, the resulting compound can clarify drug mechanisms, slow drug metabolism, and increase activity. It also improves the stability of enantiomers, inhibits configuration conversion, and reduces drug toxicity.
Dual-Tasked C-H Methylation: A Groundbreaking Strategy
Traditional methylation methods are limited to single methylation, and introducing deuterated methyl groups requires expensive reagents. To address these challenges, Professor Zhou Qianghui of Wuhan University developed an innovative Dual-Tasked C-H Methylation strategy based on the Catellani reaction [1][1]
